To use the template that edited under Setup > Accounting > Customized Fulfillment email, make sure that Use Web Site Template for Fulfillment Emails option is disabled in Setup > Accounting > Accounting Preferences > Order Management tab > Fulfillment section.
If this option is marked, it will use the Email Template in Setup > Site Builder > Set Up Web Site > Email Tab > Order Emails > Order Fulfilled section.
To test the template, edit an existing customer record with an item fulfillment transaction then change the email address, after which, edit the item fulfillment transaction then click on the Resend Order Fulfillment Email. This will send a copy of the Order Fulfillment email.
Note that "Resend Order Fulfillment Email" is a standard field that becomes available only when "Send Order Fulfilled Confirmation Emails" preference is turned on under Setup > Accounting > Accounting Preferences > Order Management tab > Fulfillment section.
